Improving Neck and Jawline Aesthetics With OnabotulinumtoxinA by Minimizing Platysma Muscle Contraction Effects: Efficacy and Safety Results in a Phase 3 Randomized, Placebo-Controlled Study

Summary: In this randomized, placebo‑controlled study, adults with moderate to severe platysma prominence (ie, visible neck bands and diminished jawline definition) received customized doses (26–36 U) of onabotulinumtoxinA and were evaluated at various time points for 120 days. By Day 14, about 77–80% of treated participants achieved at least a one‑grade improvement (versus ~22% with placebo) and roughly 41% had a two‑grade improvement (versus ~3–4% with placebo), as judged by both investigators and the patients themselves (P < .0001). These superior responder rates persisted above placebo through Day 120, and treated subjects reported significantly greater satisfaction, less bother from neck bands, and reduced psychosocial impact. OnabotulinumtoxinA was well tolerated and effectively enhanced jawline contour and neck appearance in all study participants.
